@#Introduction: Advanced glycation end products (AGEs) formation is due to glycation of lipids, proteins, and nucleic acids. AGEs play a vital role in Diabetes Mellitus (DM) and cardiovascular complications and is enhanced by oxidative stress. Hordeum vulgare (Barley Grass) is a cereal plant of grass family poaceae and has been reported to exhibit antidiabetic and antioxidant characteristics and prevention of cardiovascular disease. Current experiment focused on the evaluation of antioxidant and anti-advanced glycation end products formation effect of Hordeum vulgare water extract. Methods: Free radical scavenging ability of Hordeum vulgare water extract, was evaluated using DPPH, nitric oxide, hydrogen peroxide, lipid peroxidation, FRAP and metal ion chelating. Total phenolic content of Hordeum vulgare was also analysed. In-vitro antiglycation activity was done using BSA-MGO, BSA-Glu. Phytochemical screening was performed to detect the presence of flavonoids, saponins, tannins, steroids and terpenoids. Results: The results showed Hordeum vulgare water extract ameliorated the formation of AGE and scavenge free radical from DPPH, Nitric oxide, hydrogen peroxide, Lipid peroxidation, FRAP and Metal ion chelating. Phytochemicals such as saponins, tannins, flavonoids, steroids and terpenoids were detected in Hordeum vulgare water extract. Conclusion: Hordeum vulgare water extract reveals antiglycation and free scavenging properties.

ABSTRACT

OBJECTIVE:To study the h ypoglycemic effect of Hordeum vulgare polysaccharide(HVP)on diabetes mellitus model mice and its mechanism. METHODS :The mice were given intraperitoneal injection of streptozotocin (120 mg/kg)to induce diabetes mellitus model. After modeling ,the mice were randomly divided into model group ,metformin group (positive control , 200 mg/kg),HVP high-dose ,medium-dose and low-dose groups (300,150 and 75 mg/kg),with 10 mice in each group. Blank control group was established additionally. Administration groups were given relevant medicine intragastrically ;blank control group and model group were given constant volume of water intragastrically ,once a day ,for consecutive 30 days. The levels of fasting blood glucose (FBG)were determined after 10,20,30 days of administration. After last FBG test ,mice were intraperitoneally injected with 10% glucose solution (2 g/kg),then the area under the glucose tolerance (GTT)curve was measured at 30 and 120 min after injection. The serum levels of insulin ,the content of superoxide dismutase (SOD),glutathione peroxidase (GSH-Px), malondialdehyde(MDA)in liver tissue were detected. Pancreatic morphology of mice were detected by HE staining. RESULTS : Compared with blank control group ,the FBG ,area under GTT curve and MDA contnet in liver tissue were increased significantly in model group (P<0.01),while serum levels of insulin ,SOD and GSH-Px contents in liver tissue were decreased significantly (P<0.01);the pancreatic tissue was seriously damaged with incomplete morphology and obvious vacuoles. ABSTRACT

Abstract Pyrenophora teres f. maculata is the causal agent of barley spot form net blotch (SFNB), a major stubble-borne disease in many barley-growing areas worldwide. In plants, the Nucleotide-Binding Site-Leucine-Rich Repeat (NBS-LRR) gene family functions in immunity against a variety of pathogens and pests. From a pre-established set of NBS-type resistance gene candidates, we have selected three candidate genes, HvNBS10, HvNBS72 and HvNBS85, to analyze their possible involvement in P. teres f. maculata resistance. The studied genes were mapped on chromosomes 5H and 7H. Expression profiles using qRT-PCR, 48 hours after infection by P. teres. f. maculata, revealed that the transcription of all genes acted in the same direction (down-regulation) in both resistant and susceptible cultivars, although they showed a variation in transcript dosage. This result suggests that coordinated transcriptional responses of multiple barley NBS genes would be required to an efficient response against P. teres f. maculata. Moreover, the phylogenetic analysis revealed that the studied barley candidate R genes were characterized by a high homology with the barley Nbs2-Rdg2a gene conferring resistance to the fungus Pyrenophora graminea, suggesting a common origin of P. graminea and P. teres resistance genes in barley, following pathogens evolution. The genes characterized in the present study hold potential in elucidating the molecular pathways and developing novel markers associated with SFNB resistance in barley.

ABSTRACT

PURPOSE: Barely sprout is a well-known oriental herbal medicine with a wide range of health benefits. Recent studies have provided scientific evidence of its therapeutic effects with expanded application. This study investigated anti-melanogenic effect of barley sprout water extract (BSE) in murine melanocyte B16F10. METHODS: Various concentrations (0, 50, 125, and 250 µg/mL) of BSE and arbutin (150 ppm) were applied to B16F10 stimulated with or without alpha-melanocyte stimulating hormone (100 nM) for 72 hours. The whitening potency of BSE was determined altered cellular melanin contents. Activity and expression of tyrosinase and microphthalmia-associated transcription factor (MITF) were also assayed. RESULTS: Experimental results revealed that treatment with BSE reduced cellular melanin production by approximately 40% compared to the control. Molecular findings supported that suppressed activity and expression of tyrosinase and MITF proteins by BSE were associated with declined cellular melanogenesis. Furthermore, anti-melanogenic effect of BSE (250 µg/mL) was similar to that of arbutin, a commonly used whitening agent. Lastly, polyphenols including p-coumaric, ferulic, and vanillic acids were identified in BSE using HPLC analyses. They might be potential active ingredients showing such melanogenesis-reducing effect. CONCLUSION: BSE was evident to possess favorable anti-melanogenic potency in an in vitro model. As a natural food sourced material, BSE could be an effective depigmentation agent with potential application in pharmaceutical and cosmetic industries.

ABSTRACT

Background: The enzymes utilized in the process of beer production are generally sensitive to higher temperatures. About 60% of them are deactivated in drying the malt that limits the utilization of starting material in the fermentation process. Gene transfer from thermophilic bacteria is a promising tool for producing barley grains harboring thermotolerant enzymes. Results: Gene for α-amylase from hydrothermal Thermococcus, optimally active at 75­85°C and pH between 5.0 and 5.5, was adapted in silico to barley codon usage. The corresponding sequence was put under control of the endosperm-specific promoter 1Dx5 and after synthesis and cloning transferred into barley by biolistics. In addition to model cultivar Golden Promise we transformed three Slovak barley cultivars Pribina, Levan and Nitran, and transgenic plants were obtained. Expression of the ~50 kDa active recombinant enzyme in grains of cvs. Pribina and Nitran resulted in retaining up to 9.39% of enzyme activity upon heating to 75°C, which is more than 4 times higher compared to non-transgenic controls. In the model cv. Golden Promise the grain α-amylase activity upon heating was above 9% either, however, the effects of the introduced enzyme were less pronounced (only 1.22 fold difference compared with non-transgenic barley). Conclusions: Expression of the synthetic gene in barley enhanced the residual α-amylase activity in grains at high temperatures.

Abstract The barley HvAACT1 gene codes for a citrate transporter associated with tolerance to acidic soil. In this report, we describe a single nucleotide polymorphism (SNP) in the HvAACT1 coding region that was detected as T-1,198 (in genotypes with lower root growth on acidic soil) or G-1,198 (greater root growth) and resulted in a single amino acid change (L/V-172). Molecular dynamic analysis predicted that HvAACT1 proteins with L or V-172 were stable, although the substitution led to structural changes within the protein. To evaluate the effect of the SNP on tolerance to acidic soil, barley accessions were separated into haplotypes based on the presence of a 1 kb insertion in the HvAACT1 promoter and a 21 bp insertion/deletion. These markers and the SNP-1,198 allowed the identification of five haplotypes. Short-term soil experiments showed no difference in root growth for most of the accessions containing the 21 bp insertion and T or G-1,198. In contrast, genotypes showing both the 21 bp deletion and G-1,198, with one of them having the 1 kb insertion, showed greater root growth. These results indicate that the SNP was not advantageous or deleterious when genotypes from the same haplotype were compared. The occurrence of the SNP was highly correlated with the 21 bp insertion/deletion that, together with the 1 kb insertion, explained most of the barley tolerance to acidic soil.

ABSTRACT Recently, researches have shown that the Brazilian savannah has a great potential to supply the demand for barley grains. The purpose of this study was to assess the genetic variability in 39 elite barley (Hordeum vulgare L.) genotypes based on the agro-morphological traits of a crop irrigated in the savannah system. An irrigation experiment in the design of complete randomized block with four replicates was conducted at Federal District - Brazil. The evaluated traits were: distance from the last knot to the rachis, distance from the flag leaf to rachis, spike length, number of grains by ear, flag leaf area, plant height, silking, lodging, grain yield, thousand-seed weight, protein content and grain commercial classification. After using analysis of variance the means were used to estimate the genetic dissimilarity among all genotypes pairs based on the Mahalanobis' generalized distance. Cluster analysis using genetic distance matrix was performed having Unweighted Pair Group Method using Arithmetic Means method (UPGMA) as the criteria. Highly significant differences were found among the genotypes for all traits evaluated. The high coefficient of genetic variation indicates the possibility of having genetic gains for all traits. The traits that most contributed to the variability were the flag leaf area and silking, while the protein content and lodging were the traits that contributed the least. Based on the cluster analysis, at least three major groups of similarity were found. There was a clustering trend of two and six-rowed materials. The most divergent genotypes were PFC 2005123, Antártica-1, Nandi and FM 404.


RESUMO Recentemente, pesquisas tem demonstrado que o Cerrado tem grande potencial para suprir a demanda de cevada. O objetivo deste trabalho foi avaliar a variabilidade genética de 39 genótipos elite de cevada com base em características morfoagronômicas avaliadas em sistema de produção irrigado no Cerrado. O experimento foi conduzido num delineamento de blocos ao acaso com quatro repetições no Distrito Federal - Brasil. Avaliaram-se as características: distância do último nó à ráquis, distância da folha bandeira à ráquis, comprimento da espiga, número de grãos por espiga, área da folha bandeira, altura de plantas, espigamento, grau de acamamento, rendimento de grãos, peso de mil sementes, teor de proteína e classificação comercial de grãos. Após análise de variância as médias foram utilizadas para estimar a dissimilaridade genética, com base na distância generalizada de Mahalanobis. Utilizando a matriz de distâncias genéticas foram realizadas análises de agrupamento. Foram observadas diferenças altamente significativas entre os genótipos para todas as características avaliadas. O elevado coeficiente de variação genético evidencia a possibilidade de obter ganhos genéticos para todas características. Através das análises de agrupamento, verificou-se a formação de pelo menos três grandes grupos de similaridade. Houve tendência de agrupamento dos materiais dísticos e hexásticos. Os genótipos PFC 2005123, Antártica-1, Nandi e FM 404 foram os mais divergentes.

ABSTRACT

Objetivou-se com o trabalho avaliar os efeitos de épocas de emergência das plantas daninhas, azevém e nabo, sobre a cultura da cevada. O experimento foi conduzido em delineamento de blocos casualizados, arranjado em esquema fatorial 2x5, com três repetições. No fator A, foram alocadas as plantas competidoras (azevém e nabo) e no B as épocas de emergência (14 e 07 dias antes, no mesmo dia, 07 e 14 dias depois da emergência da cultura). No início da floração da cevada, foram mensuradas as variáveis: estatura das plantas, número de colmos ou caules, área foliar e matéria seca da parte aérea da cultura e das plantas daninhas. Todas as variáveis avaliadas da cevada apresentaram aumento de valor com o atraso da emergência das plantas daninhas. As espécies, cultivada ou daninhas, que emergiram antes, apresentaram maior habilidade competitiva, dominando o ambiente quando emergiram 14 dias antes uma da outra. Quanto mais tarde ocorrer a emergência das plantas daninhas com relação à cultura, menor é a interferência e, desse modo, maior é a habilidade da cevada em competir com a espécie daninha pelos recursos disponíveis no meio.


The objective was to evaluate the effects of times of emergence of the weed ryegrass species and wild radish on barley. The experiment was conducted in a randomized block design, arranged in a 2x5 factorial design, with three replicates. In factor A were allocated the competing species (ryegrass and wild radish) and in factor B the emergence periods (14 and 7 days before, on the same day, 7 and 14 days after crop emergence). At the beginning of barley flowering, the following variables were quantified: plant height, number of stalks or stems, leaf area, and shoot dry mass of crop and weeds. All barley variables had increased in value with the delay in weed emergence. The species, cultivated or weeds, that emerged earlier showed greater competitive ability, dominating the environment when emerging 14 days before each other. The later the emergence of weeds with respect to the crop, the less the interference they cause, and, thereby, the crop presents a greater ability to compete with the weeds for available resources in the environment.

Background In the present study populations, representing different rounds of recombination were used for the analysis of phenotypic effects associated with the sdw1/denso locus. Other studies have mostly focused only on one type of population. Many different QTLs mapped at the same position as the sdw1/denso locus may indicate a pleiotropy of this gene or a tight linkage between genes conditioning quantitative traits. To date, results of studies have not unequivocally proven either of these two phenomena. Results Both breeding and molecular mapping experiments were undertaken to examine 200 single seed descent (SSD) and 60 doubled haploid (DH) lines obtained from the Maresi (with a semi-dwarfing gene) and Pomo cross combination. They were evaluated for the type of juvenile growth habit and certain agronomic traits were measured after harvesting. The estimates of mean values, standard errors and significance of effects were analyzed. In terms of the analyzed characteristics, the greatest variability was obtained for genotypes with the prostrate growth habit. Microsatellite markers (SSR) were also used to identify co-segregation with the sdw1/denso locus and Bmag0013, Bmag0877, Bmag0306b markers were linked the closest. A partial linkage map of chromosome 3H with the sdw1/denso semi-dwarfing gene was constructed and QTLs were identified. Conclusions Our experiments confirmed the impact of the semi-dwarfing gene on plant height, heading and flowering date both in SSD and DH populations, which may indicate pleiotropy. Moreover, a partial linkage between sdw1/denso locus and grain weight per spike and 1000-grain weight was found in the SSD population.

The objective of this work was to characterize and quantify the genetic variability of 39 barley elite genotypes from a Brazilian working collection belonging to Embrapa, using RAPD (Random Amplified Polymorphic DNA) molecular markers. Genomic DNA samples were extracted from leaves of each genotype and 15 decamer primers were used to obtain RAPD molecular markers. Molecular markers were converted in a binary data matrix utilized to estimate genetic dissimilarities between genotypes and to realize grouping and dispersion graphic analysis. A total of 160 RAPD markers were obtained, making 10.7 markers medium per primer. From all the markers, 141 (88.12%) were polymorphic. Genetic dissimilarities varied from 0.049 to 0.337 among the genotypes. PFC 2004033 and Prestige cultivar showed biggest genetic dissimilarities to others genetic materials. Grouping and dispersion graphic analysis showed a clustering tendency between the Mexican and American genotypes. Another clustering tendency was also found concerning the six-rowed materials. Accessions developed and used in Brazil and also in Germany, UK and Australia have shown the greatest genetic dissimilarity among themselves, being considered promising options to increase the genetic base of breeding programs.


O objetivo deste trabalho foi caracterizar e quantificar a variabilidade genética de 39 acessos de cevada elite da coleção de trabalho da Embrapa Cerrados, utilizando marcadores moleculares RAPD. Foram utilizados 15 iniciadores decâmeros para a obtenção dos marcadores RAPD, que foram convertidos em uma matriz de dados binários, a partir da qual foram estimadas as dissimilaridades genéticas entre os diferentes acessos e realizadas análises de agrupamento. Foram obtidos 160 marcadores RAPD, dos quais 141 (88,12%) foram polimórficos. As dissimilaridades genéticas variaram de 0,049 a 0,337, entre os acessos de cevada. A análise de agrupamento e de dispersão gráfica mostrou uma tendência de agrupamento entre os genótipos mexicanos e americanos. Outra tendência de agrupamento também foi encontrada entre os genótipos de seis fileiras de grãos. Acessos desenvolvidos e utilizados no Brasil e também os genótipos provenientes da Alemanha, Inglaterra e Austrália têm demonstrado a maior divergência genética entre si, sendo considerados opções interessantes para aumentar a base genética dos programas de melhoramento.

Barley (Hordeum vulgaris L.) is the second most important winter crop in Southern Brazil. The excessive rainfall in this region during the crop-growing season increases the frequency and intensity of foliar fungal diseases. The research aimed to determine the damage function equations (DFE) for the multiple pathosystem of barley brown spot and powdery mildew based on the relationship between grain yield and diseases intensity at different 'BRS Cauê' cultivar growth stages (GS) during 2009 and 2010 growing seasons in Southern Brazil. The experiments were arranged in a randomized complete block design with nine treatments and four replicates. The disease gradients were generated by strobilurins and triazols fungicides rates and number of applications on barley cv. Cauê. The fungicide applications and disease incidence and severity assessments were performed at the 22, 31, 39, 45 and 56 plant GS. The DFE were obtained by variance analysis and linear regression between grain yield and diseases intensity. Significant and negative DFE were obtained and the damage coefficients (DC) varied from 29.48 to 100.08 (2009) and from 36.08 to 113.57kg ha-1 (2010) for incidence, and from 219.5 to 6,276.6 (2009) and 102.3 to 5,292.5kg ha-1 (2010) for severity. The largest damage coefficients were obtained when diseases assessments were made on GS 22 and 31 on both growing seasons evaluated. DFE were used to calculate the economic damage threshold (EDT) as a criterion to indicate the fungicide application moment to control the diseases in cultivars similar to 'BRS Cauê' in Southern Brazil.


A cevada (Hordeum vulgaris L.) é a segunda mais importante cultura de inverno no Sul do Brasil. Nessa região, o excesso de chuvas durante a estação de crescimento da cultura favorece o aumento na frequência e intensidade de doenças foliares. O trabalho objetivou determinar as equações de função de dano (EFD) para o patossistema múltiplo mancha-marrom e oídio da cevada pela relação entre rendimento de grãos e intensidade foliar das doenças em diferentes estádios fenológicos da cultivar de cevada 'BRS Cauê', nas safras 2009 e 2010 no sul do Brasil. O delineamento foi em blocos casualizados com nove tratamentos e quatro repetições. Os tratamentos foram constituídos de diferentes doses e número de aplicações dos fungicidas triazóis e estrobilurinas para gerar os gradientes de intensidade das doenças. As aplicações de fungicidas e as avaliações da incidência e severidade foliar foram realizadas nos estádios fenológicos (EF) 22, 31, 39, 45 e 56. As equações foram obtidas pela análise de variância e regressão linear entre rendimento de grãos e intensidade das doenças. As EFD foram significativas e negativas e os coeficientes de danos variaram de 29,48 a 100,08kg ha-1 (2009) e 36,08 a 113,57kg ha-1 ( 2010) para a incidência e de 219,5 a 6.276,6kg ha-1 ( 2009) e 102,3 a 5.292,5kg ha-1 (2010). Os maiores coeficientes de dano foram obtidos nos estádios EF 22 e 31em ambas as safras. As EFD foram usadas para calcular o limiar de dano econômico, que é um critério que indica o correto momento de aplicação de fungicidas para o controle das doenças em cultivares similares a 'BRS Cauê' no sul do Brasil.

Objetivou-se quantificar os compostos fenólicos em extrato hidroetanólico de grãos das cultivares de cevada ('BRS Lagoa' e 'MN 743') em dois anos consecutivos de cultivo (2005 e 2006), avaliando a influência dos fatores climáticos nesse conteúdo. Os compostos fenólicos totais foram quantificados pelo método de Folin-Ciocalteau e os teores de rutina e ácidos cafeico e ferúlico por CLAE em fase reversa. O conteúdo de compostos fenólicos totais variou de 0,97 a 1,59 mg AG g-1 de cevada e os fatores climáticos durante o período de plantio e colheita influenciaram no conteúdo. Foram identificados e quantificados nessas cultivares rutina, ácido cafeico e ácido ferúlico. A avaliação dos fatores climáticos no conteúdo de compostos fenólicos em cevada é de grande interesse, visando a sua importância nutricional e recomendação de cultivares com conteúdos expressivos de compostos bioativos.


This study aimed to quantify the phenolic compounds in hydroethanolic extract of cultivar seeds ('BRS Lagoa' and 'MN 743') in two consecutive years of cultivation (2005 and 2006) evaluating the influence of climatic factors in such content. The total phenolic compounds were quantified by Folin-Ciocalteau's method and the levels of rutin and caffeic and ferulic acids by HPLC in the reversed-phase. The concentration of total phenolic compounds ranged from 0.97 to 1.59 mg GA g-1 of barley and the climatic factors during the planting and harvesting influenced the barley content. Caffeic, rutin and ferulic acid were identified and quantified in these cultivars. Algumas características relacionadas à qualidade do malte podem ser trabalhadas não só ao nível genético e/ou como respostas de interações ambientais, mas também ao nível de processo industrial. O objetivo do presente trabalho foi avaliar o efeito da temperatura sobre a atividade das β-glucanases e degradacão de β-glucanos durante os quatro dias finais de germinação do processo de malteação. Quatro cultivares de boa qualidade de malteação ('N721', 'N743', 'Scarlett' e 'Sebastian') foram analisadas e os resultados obtidos demonstraram que elas tiveram desempenho diferente quanto à atividade enzimática e à degradação de β-glucanos nas diferentes temperaturas testadas. A utilização de temperatura um pouco mais elevada no início da germinação (22°C) foi a mais efetiva na produção das variáveis estudadas. Para a cultivar 'N721', constatou-se o melhor desempenho comparativamente às demais, indicando um padrão mais homogêneo de degradação de β-glucanos durante a germinação e também maior eficiência na degradação desde as 96 horas do processo. Alta atividade das β-glucanases e redução no tempo de malteação são de extremo interesse para a indústria malteira, pois representa uma otimização no volume de malte produzido e consequentemente uma redução de custos para a sua produção.


Some characteristics related to the quality of malt can be worked not only at the genetic level and/or as responses to environmental interactions, but also in industrial process. This study assesses the effect of temperature on β-glucanase activity and on the degradation of β-glucans during the last four days of germination, in the malting process. Four cultivars of good quality malting ('N721', 'N743', 'Scarlett' and 'Sebastian') were analyzed and the results showed they had different performance on the enzyme activity and β-glucan degradation, at the different temperatures tested. A slightly higher temperature at the beginning of germination (22°C) was more effective in terms of the variables studied. Cultivar 'N721' presented the best performance compared to other cultivars, revealing a more homogeneous pattern of β-glucan degradation during germination and also increased efficiency in degrading -glucans as early as in the 96th hour of the germination process. High activity of β-glucanases and shorter time is very interesting to the brewery industry, since it represents an optimization of the malt volume produced and a cost reduction for its production.

Populações duplo-haplóides apresentam especial vantagem para análises genéticas, uma vez que a informação que elas oferecem pode ser maximizada, devido ao fato que todos os locos encontram-se em homozigose. O objetivo deste trabalho foi o desenvolvimento de duas populações duplo-haplóides (DHs) de cevada (Hordeum vulgare ssp. vulgare L.) segregantes para a atividade das enzimas (1-3, 1-4)-β-glucanases, através da técnica de cultura de anteras. Foram realizados dois cruzamentos com cultivares contrastantes para esta característica. As cultivares parentais selecionadas foram 'MN 698' e 'CEV 97047', para o desenvolvimento da população "malte verde" (MV), e 'Embrapa 127' e 'CEV 96025' para o desenvolvimento da população "malte seco" (MS). Foram cultivadas 10.734 anteras da população MS e 4.139 anteras da população MV. A população MV produziu 50 por cento mais plântulas verdes quando comparada à população MS, refletindo a importância do genótipo na resposta à cultura de anteras e na regeneração. A maioria das plantas adultas duplo-haplóides foi obtida através da duplicação espontânea in vitro do genoma haplóide, ocorrendo em 66 por cento das plantas da população MS e 76 por cento das plantas da população MV. Também foram observadas, em menor frequência, plantas haplóides, triplóides e tetraplóides. Através da técnica de cultura de anteras, foram desenvolvidas 204 linhagens duplo-haplóides, das quais 72 linhagens são da população "malte seco" e 132 linhagens são da população "malte verde". Este material constitui um importante germoplasma para o melhoramento genético da cevada.


Doubled haploid populations offer special advantages in genetic analyses, since the information they provide may be maximized due to the fact that all loci are homozygous. The aim of this study was to develop two barley (Hordeum vulgare ssp.vulgare L.) doubled-haploid (DHs) populations segregating to (1-3, 1-4)-β-glucanases activity, utilizing the anther culture protocol. Two crosses were performed with contrasting cultivars to (1-3, 1-4)-β-glucanases activity. Parental cultivars used were 'MN 698' and 'CEV 97047' for the development of 'green malt' population (MV) and Embrapa 127 and 'CEV 96025' for the development of "dry malt" population (MS). For the MS and MV populations, 10,734 and 4,139 anthers were cultured, respectively. MV population produced 50 percent more green seedlings as compared to MS population, which reflects the importance of genotype to the culturing of anthers and to regeneration. Most doubled-haploid adult plants were obtained by in vitro spontaneous duplication of the haploid genome, which occurred in 66 percent of plants from the MS population and 76 percent of plants from the MV population. Haploid, triploid and tetraploid individuals were also observed, though at low frequencies. The anther culture protocol afforded to develop 204 double-haploid lineages, of which 72 were generated by the 'dry malt' population and 132 from the "green malt" population. This material should be considered as important germplasm for barley genetic improvement.

Genetic diversity in 403 morphologically distinct landraces of barley (Hordeum vulgare L. subsp. vulgare) originating from seven geographical zones of Asia was studied using simple sequence repeat (SSR) markers from regions of medium to high recombination in the barley genome. The seven polymorphic SSR markers representing each of the chromosomes chosen for the study revealed a high level of allelic diversity among the landraces. Genetic richness was highest in those from India, followed by Pakistan while it was lowest for Uzbekistan and Turkmenistan. Out of the 50 alleles detected, 15 were unique to a geographic region. Genetic diversity was highest for landraces from Pakistan (0.70 ± 0.06) and lowest for those from Uzbekistan (0.18 ± 0.17). Likewise, polymorphic information content (PIC) was highest for Pakistan (0.67 ± 0.06) and lowest for Uzbekistan (0.15 ± 0.17). Diversity among groups was 40 percent compared to 60 percent within groups. Principal component analysis clustered the barley landraces into three groups to predict their domestication patterns. In total 51.58 percent of the variation was explained by the first two principal components of the barley germplasm. Pakistan landraces were clustered separately from those of India, Iran, Nepal and Iraq, whereas those from Turkmenistan and Uzbekistan were clustered together into a separate group.

Se realizaron dos experimentos en condiciones de campo en los ciclos de invierno-primavera (ECIP) bajo riego y verano (ECV) bajo temporal, y un experimento en invernadero ciclo verano (EIV), para estudiar el efecto de las condiciones del ambiente en el rendimiento de grano (RG) y calidad de la semilla en cebada y trigo. En un análisis combinado de los experimentos de campo la cebada expresó mayor RG, materia seca de la parte aérea (MSPA), número de espigas/m² y número de granos/m² (NGPMC) en 21, 31, 28 y 22% respectivamente respecto a trigo; los genotipos de cebada fueron 6cm más altas y alcanzaron su mayor rendimiento 18 días antes que los genotipos de trigo; aunque el trigo mostró 11% mayor IC que cebada. El RG, índice de cosecha (IC), MSPA y NGPMC y altura de planta disminuyeron para cebada en 30, 29, 2,12% y 25cm y para trigo en 38, 10, 32, 27% y 32cm respectivamente, al pasar de riego a temporal; el número de días a antesis en cebada ocurrió 28 días y en trigo 46 días antes en temporal que en riego. También en EIV la cebada fue superior sobre trigo en el RG y MSPA en 17 y 33, respectivamente. La cebada presentó una mayor calidad de la semilla que los genotipos de trigo en los parámetros de vigor de las plántulas, mientras que los materiales de trigo presentaron 10 y 23% mayor germinación y peso volumétrico, respectivamente.


Two experiments were carried out under field conditions; one was conducted in winter-spring (FEWS) growing season under irrigation and another experiment was conducted under rainfall conditions in the summer (FES), while a third experiment was carried out in green house conditions in the summer (GES), in order to study the environmental effects in grain yield (GY) and seed quality between barley and bread wheat. In the combined analysis of the experiments carried out under field conditions, the results showed that the barley had 21, 31, 28 y 22% higher grain yield (GY), above-ground dry matter (AGDM), number of spikes/m², number of grains/m² (NGPSM), respectively, with respect to wheat. Barley plantlets were 6cm higher and reached its larger yield 18 days before those of wheat, although wheat showed an 11% larger harvest index (HI) than barley. GY, HI, AGDM, NGPSM and plant height all decrease in FES for barley in 30, 29, 2, 12% and 25cm, respectively, and for wheat in 38, 10, 32, 27% and 32cm, respectively, compare with FEWS. The number of days to anthesis was 28 days and 46 days before in FES than in FEWS, respectively, in barley and wheat. Also, in GES barley was better than the wheat on GY and AGDM in 17 and 33%, respectively. Barley showed a better seed quality than wheat in plantlet vigor, whereas wheat plantlets showed 10 and 23% larger germination and volumetric weight, respectively.

Em uma análise combinada dos experimentos de campo a cevada expressou maior RG, matéria seca da parte aérea (MSPA), número de espigas/m² e número de grãos/m² (NGPMC) em 21, 31, 28 e 22% respectivamente em relação ao trigo; os genótipos de cevada foram 6cm mais altos e alcançaram seu maior rendimento 18 días antes que os genótipos de trigo; embora o trigo tenha mostrado 11% maior Índice de colheita (IC) que a cevada. O RG, índice de colheita (IC), MSPA e NGPMC e altura de planta diminuiram para cevada em 30, 29, 2 12% e 25cm e para trigo em 38, 10, 32, 27% e 32cm respectivamente, ao passar de irrigação a temporal; a antese em cevada ocurreu em 28 dias e no trigo em 46 dias mais cedo em temporal do que em irrigação. Também em EEV a cevada foi superior sobre trigo no RG e MSPA em 17 e 33, respectivamente. A cevada apresentou uma maior qualidade da semente que os genótipos de trigo nos parâmetros de vigor das plântulas, enquanto que os materiais de trigo apresentaram 10 e 23% maior germinação e peso volumétrico, respectivamente.

ABSTRACT

Molecular genetic research relies heavily on the ability to detect polymorphisms in DNA. Single nucleotide polymorphisms (SNPs) are the most frequent form of DNA variation in the genome. In combination with a PCR assay, the corresponding SNP can be analyzed as a derived cleaved amplified polymorphic sequence (dCAPS) marker. The dCAPS method exploits the well-known specificity of a restriction endonuclease for its recognition site and can be used to virtually detect any SNP. Here, we describe the use of the dCAPS method for detecting single-nucleotide changes by means of a barley EST, CK569932, PCR-based marker.

ABSTRACT

The development of in vitro haploid plants followed by spontaneous or induced genome duplication allows to achieve, in one generation, the recovery of total homozygosis. The efficiency of the haplodiploidization process through in vitro anther culture of barley is variable among genotypes. This study was aimed at determining the androgenetic response of nine barley genotypes from the breeding program of Embrapa Trigo, analysing proembryoid development and green plantlets regeneration in anthers cultivated in vitro. Cultivar 'BR2' presented the highest average of proembryoids (104/anther) and 'MN698' presented the highest average of green plantlets (0,41/anther). There was a significant variation among the average values of barley genotypes for embryo formation and green plantlets regeneration, making possible the selection to combine androgenetic capacity and good agronomic traits.


A obtenção, na cevada, de plantas haplóides in vitro e a posterior duplicação natural ou artificial do genoma permitem alcançar a homozigose completa, em uma geração. A eficiência da haplodiploidização pela cultura de anteras é variável entre os genótipos. Foi avaliada a resposta androgenética através da formação de pró-embrióides e da regeneração de plântulas verdes em nove cultivares do programa de melhoramento de cevada da Embrapa Trigo, em anteras cultivadas in vitro. A cultivar "BR2" apresentou maior média de pró-embrióides (104/antera), enquanto "MN698" mostrou a maior média de plântulas verdes (0,41/antera). Houve variação significativa entre os valores médios dos genótipos em relação à formação de pró-embrióides e à regeneração de plântulas verdes, indicando a possibilidade de seleção para combinar a capacidade androgenética com boas características agronômicas.

O presente trabalho teve como objetivo avaliar o teor de proteínas em grãos de cevada em resposta à aplicação de N em estádios de desenvolvimento da cultura. Os experimentos foram conduzidos em Eldorado do Sul e Encruzilhada do Sul, no ano de 2000, e em Victor Graeff, nos anos de 2000 e 2001, em delineamento experimental em blocos casualizados com quatro repetições. Os estádios de aplicação de N foram na emergência das plântulas; na emissão da 2ª ou 3ª folha; da 4ª ou 5ª folha; 6ª ou 7ª folha; 8ª ou 9ª folha; e no emborrachamento. As doses de N foram de 30 ou 40kg ha-1 e 60 ou 80kg ha-1, para a menor e maior dose, respectivamente. As determinações realizadas foram teor de proteínas nos grãos e número de grãos metro-2. Para os experimentos realizados em 2000, a aplicação de nitrogênio até o início do alongamento dos entrenós (emissão da 7ª folha) manteve o teor de proteínas no grão abaixo dos 12 por cento, mesmo para a maior dose de N. Os teores de proteínas no grão em Victor Graeff, no ano de 2001, ficaram acima do limite máximo de 12 por cento com a aplicação da maior dose de N já em estádios iniciais de desenvolvimento da cultura, devido ao maior teor de matéria orgânica no solo em relação aos outros locais.


This study was was aimed at evaluating the protein content in barley seeds affected by nitrogen application in different growth stages. Experiments were carried out in Eldorado do Sul and Encruzilhada do Sul, in 2000, and Victor Graeff, in 2000 and 2001, on a randomized blocks scheme with four repetitions. The growth stages of N application were in emergency of seedlings; emission of 2nd or 3rd leaf; 4th or 5th leaf; 6th or 7th leaf; 8th or 9th leaf; and in boot stage. The N rates were 30 or 40kg ha-1 and 60 or 80kg ha-1, for smallest and largest N rate, respectively. The protein content in barley seeds and the number of grains area-1 were determined. In the experiments carried out in 2000 the nitrogen application until the beginning of the internodes elongation maintained the seeds protein content below of 12 percent, inclusive in the largest N rate. The seeds protein content in Victor Graeff, 2001, surpassed the maximum limit of 12 percent with N applications in initial growth stages, decurrently of the higher soil organic matter content in relation to others experiments.

O trabalho teve como objetivos avaliar o acúmulo de matéria seca (MS) e de nitrogênio (N) da parte aérea de plantas de cevada e a taxa de sobrevivência de colmos em função da época de aplicação de N. Um experimento foi realizado em Eldorado do Sul, RS, em 2000, testando seis épocas de aplicação de N (emergência das plântulas, emissão da 3ª folha, 5ª folha, 7ª folha, 9ª folha do colmo principal (CP) e no emborrachamento) e duas doses de N (40 e 80kg ha-1). Foram determinados os acúmulos de MS e N da parte aérea no período entre o início do alongamento dos colmos e o florescimento e a taxa de sobrevivência de colmos. As aplicações de N entre a emergência das plântulas e a emissão da 9ª folha do CP resultaram nas maiores taxas de acúmulo de MS. Entretanto, as maiores taxas de acúmulo de N foram observadas com a aplicação de N no período de alongamento dos colmos. As maiores porcentagens de sobrevivência de colmos também foram observadas com a aplicação de N nesse período, mostrando a relação existente entre a necessidade de N para dar aporte ao alongamento dos colmos e a mortalidade de afilhos.


This study was aimed at evaluating the dry matter (DM) and nitrogen (N) accumulation in aerial part of barley plants and the shoot survival rate in function of time of N supply. An experiment was carried out in Eldorado do Sul, RS, in 2000, where six times of N supply (seedling emergence; 3rd leaf; 5th leaf; 7th leaf; 9th leaf emergence and boot stage) and two N rates (40 and 80kg ha-1) were tested. The DM and N accumulation among the shoot elongation begin and the bloom stage and the shoot survival rate were determined. The N supply among seedling emergence and 9th leaf emergence stage resulted the largest DM rates accumulations. However, the largest N rates accumulations were observed in shoot elongation period. The largest shoot survival rates were also observed in this period, showing the relation among the N necessity to support the shoot elongation process and the shoot mortality.
